Answer:

In the event of marking a check box for a negatively phrased option on a Constituent record, such as "Requests No Email" or "Deceased" a value of "Yes" will place a check mark in the check box. A value of "No" will have no effect.

You may place a column in your data file with a value of "Yes" on all applicable rows or you can place a virtual column on your profile itself if the check mark should be placed on all incoming records. Please be sure to check the box to allow updated to Bio1 checkbox fields.
